D-Luciferin (potassium salt)是D-Luciferin(D-萤光素)的钾盐形式,是萤火虫萤光素酶的底物,用于生物发光成像。
体内可见光成像技术主要包括生物发光(bioluminescence)与荧光(fluorescence)成像两种技术。
其中生物发光是基于萤光素酶报告基因系统中萤光素酶的发光。萤光素酶是自然界中能够产生生物荧光的酶的统称,最常见的是萤火虫萤光素酶。编码萤光素酶的基因可以在体外转染到细胞染色体DNA上以表达萤光素酶。当细胞分裂和分化时, 萤光素酶也会得到持续稳定的表达。将标记好的细胞接种到实验动物体内,当腹腔或静脉注射相应的底物后,可迅速产生发光现象。萤光素酶基因广泛用作各种细胞中的报告基因。
萤光素酶的发光不需要激发光,但需要底物。萤光素酶在ATP和氧存在的条件下,催化底物发生氧化反应才可以发光。常见的萤光素酶有两种,分别是萤火虫萤光素酶(firefly luciferase,编码基因是luc)和海肾萤光素酶(Renilla luciferase,编码基因是Rluc)。二者的底物不一样,前者的底物是D-Luciferin,后者的底物是Coelenterazine。
D-Luciferin(D-萤光素)作为萤火虫萤光素酶的底物,当被萤光素酶催化氧化时,会在560 nm处产生化学发光。被广泛用于活体小鼠的无创成像[1]。萤光素、萤光素酶和萤火虫萤光素酶也经常被称作荧光素、荧光素酶和萤火虫荧光素酶。
有机阴离子转运蛋白1(OAT1)可作为d-萤光素(d-luc)转运蛋白,用于增强萤光素酶表达细胞的生物发光(BL)强度。在表达Pyrearinus termitilluminans萤光素酶的HEK293细胞(HEK293 / eLuc)中表达OAT1,d-luc的细胞内积累增加,生物发光显著增强[2]。2-羟丙基-β-环糊精(HPCD)结合d-萤光素,减少d-萤光素的细胞积累,干扰生物发光信号[3]。生物发光成像(BLI)裸鼠的D-萤光素皮下注射(SC)是腹膜内(IP)注射的一种方便有效的替代方法,特别是对于信号较弱和/或需要更高精确度的肿瘤。在注射后7分钟内,SC和IP途径在皮下,颅内,舌头和肺异种移植肿瘤模型中产生相似的发光。IP注射有时不可见肿瘤,而SC注射可可靠地观察所有肿瘤[4]。
