在真核生物中开发了调节细胞周期蛋白依赖性激酶(CDK)的高度保守的机制阵列,CDK控制细胞周期进程和细胞分裂的时间。CDK抑制剂在细胞增殖、分化、衰老以及程序性死亡中发挥关键的调节作用。
体外:将Bohemine施用于小鼠杂交瘤细胞的培养物,以评价其对细胞生长和单克隆抗体产生的能力。结果表明,加入浓度范围为1-10 μM的bohemine导致生长和单克隆抗体产生的短期停滞。这种细胞功能的短期抑制之后是比生长速率和比产生速率的显著短时增加。在半连续培养中,稳态活细胞密度值在微摩尔浓度bohemine存在下显示出一定的细胞生长刺激,在10和30 μM bohemine存在下表现出生长抑制[1]。
体内:在小鼠中,静脉注射后首个60分钟,bohemine迅速和完全代谢并在循环中消失。此外,代谢物被肝胆管和肾脏排泄消除[2]。
临床试验:到目前为止,bohemine仍处于临床前研究阶段。
