α-1抗胰蛋白酶(A1AT)的氨基酸序列为H2N-Leu-Gln-His-Leu-Glu-Asn-Glu-Leu-Thr-His-Asp-Ile-Ile-Thr-Lys-OH,又被称为血清胰蛋白酶抑制剂,属于丝氨酸蛋白酶抑制剂超家族。
A1AT是一个单链糖蛋白,成熟状态的A1AT包含394个氨基酸,并具有一定数量的糖基化侧链。三个N-连接糖基化位点连接两触角的N型多聚糖(diantennary N-glycans)。然而,天冬酰胺107是一个特别的位点,这个位点具有特定的异质性,可以连接三触角甚至四触角N-聚糖。
A1AT是人血清中丝氨酸蛋白酶的主要抑制剂,主要产生于肝脏,也可通过肝外的细胞产生,包括中性粒细胞和某些癌细胞[1]。AAT是一种急性时相蛋白,在急性炎症中,其浓度比正常高3到4倍[2]。有几种类型的癌症,包括非小细胞肺癌,与血清中AAT的增加相关[3]。临床研究表明,AAT的高循环水平与肿瘤发生直接相关。最近有研究发现,在肺癌患者中,特别是在转移的情况下,AAT的血浆水平显著升高[4]。此外,不仅天然的抑制性形式AAT可以调节肿瘤生长和侵袭,构象修饰的非抑制形式AAT也在这个过程中发挥重要作用。Kataoka和同事已经证明,AAT的C-末端片段可增强人体内胰腺癌细胞的生长和侵袭[5]。
