AIM-100是一种小分子Ack1酪氨酸激酶抑制剂,IC50值为24 nM[1]。
AIM-100模仿ATP,且显著地、特异性抑制Ack1活性。AIM-100对其他30个激酶(包括PI3-激酶和AKT)无抑制活性,且其对Lck具有5倍更高的IC50值。在EGF处理的MEF细胞中,AIM-100显著降低Ack1活性。在人前列腺癌细胞LNCaP和LAPC4中,AIM-100用药可导致G0/G1期细胞数量增加及随后的细胞生长抑制。AIM-100的这些效果对胰腺癌细胞也同样起作用。AIM-100在10 μM浓度时诱发Panc-1细胞的凋亡。此外,AIM-100抑制细胞生长,在CD-18、Panc-1、OV90、MCF-7及MDA-MB-468癌细胞中的GI50值在7到8 μM范围内[1, 2]。
