6-thio-dG是核苷类似物[1],是端粒酶介导的端粒扰乱化合物[2]。它是一种抗肿瘤剂[1]。癌症细胞对6-thio-dG非常敏感,根据细胞类型,IC50值范围为0.7-2.9 μM[3]。
端粒存在于真核细胞的线性染色体末端。它们对基因组稳定性和染色体维持很重要[3]。
在HCT116人结肠癌细胞系中,6-thio-dG促使独立于端粒酶活性的端粒缩短,诱导端粒功能障碍。GRN163L是一种端粒酶抑制剂。在HCT116细胞中, GRN163L和6-thio-dG共同使用会增加端粒缩短。1周内,6-thio-dG杀死大多数HCT116细胞,改变细胞形态。正常BJ成纤维细胞端粒酶处于沉默状态。6-thio-dG处理1周后,显示对细胞形态没有影响。6-thio-dG长期处理对端粒缩短无影响[1]。
在异种移植A549肺癌细胞的小鼠模型中,与对照相比,腹膜内隔日注射2 mg/kg的6-thio-dG,完全阻止了肿瘤生长。Ki67是与增殖水平相关的生物标志物。与对照组相比,6-thio-dG降低了Ki67染色。与未处理的对照相比,局部注射6-thio-dG显著降低了肿瘤生长速率 [3]。
