表皮生长因子(Epidermal Growth Factor, EGF)最初是在从小鼠颌下腺制备的神经生长因子的粗制剂中发现的，当注射到新生小鼠体内时，表皮生长因子会引起早期眼睑张开、门牙出疹、毛发生长抑制和生长发育迟缓。从尿中分离到人EGF，根据其抑制胃液分泌的作用，命名为尿胃泌素。EGF是来源于膜锚定前体的生长因子家族的原型。该家族的所有成员的特点是在其细胞外结构域存在至少一个EGF结构单元(由形成三个二硫键的保守6半胱氨酸基序的存在来定义)。EGF最初作为130 kDa的前体跨膜蛋白合成，含有9个EGF单位。成熟的可溶性EGF序列对应于位于跨膜结构域近端的EGF单元。膜EGF前体能够与EGF受体结合，据报道具有生物活性。成熟的人EGF与成熟的小鼠和大鼠EGF具有70%的a.a.序列一致性。
