HPF是一种细胞可渗透的,光稳定的羟苯基荧光素,可作为检测高活性氧(hROS)的高度灵敏的荧光探针[1]。有许多荧光试剂,例如2,7-二氯二氢荧光素(DCDHF),可用于检测自由基,但是它们容易被光氧化以及许多非自由基氧化剂(例如过氧化氢)氧化,因此存在很大的局限性。而HPF仅被高活性氧(hROS)氧化,例如氢氧自由基,过氧亚硝酸盐和过氧化物酶/ H2O2系统产生的hROS。该化合物对其他活性氧(ROS),例如次氯酸盐,单线态氧,超氧化物,过氧化氢,一氧化氮和烷基过氧化物几乎没有活性。
HPF本身没有荧光,当与hROS(如羟基自由基和过亚硝酸盐)反应被氧化后,HPF转换为高荧光分子荧光素,激发/发射最大值分别为490/515 nm。
由于HPF不会与一氧化氮,超氧化物和过氧化氢反应,因此可以与一氧化氮和超氧化物区分开来检测亚硝酸盐。由于HPF不与次氯酸盐(-OCl)反应,因此可以与检测-OCl的APF(氨基苯基荧光素)结合使用[2,3]。
