表皮生长因子受体(EGFR)存在于细胞膜上,特异性配体结合激活受体分子,配体包括表皮生长因子和转化生长因子α。EGFR的二聚化刺激其细胞内固有的蛋白酪氨酸激酶活性,导致EGFR的C-末端几个酪氨酸(Y)残基上发生自磷酸化,包括Y992、Y1045、Y1068、Y1148和Y1173。这个自磷酸化导致下游信号转导的激活,下游信号通路蛋白主要包括可以通过自身SH2结构域与磷酸化的酪氨酸结合的蛋白质。这些下游信号蛋白启动多个级联信号传导途径,主要是MAPK、Akt和JNK途径,从而导致DNA合成和细胞增殖。这类蛋白质参与调节细胞过程,例如细胞迁移、粘附和增殖。受体的激活对于人体皮肤的先天免疫应答非常重要。
许多治疗方法靶向表皮生长因子受体。单克隆抗体阻断了胞外配体结合的结构域。结合位点被阻断,导致信号分子不能结合,不能激活酪氨酸激酶。另一种治疗方法是使用小分子物质来抑制受体的胞内EGFR酪氨酸激酶。失去激酶活性,EGFR无法激活自身,导致下游接头蛋白失去结合的先决条件,阻止细胞内的信号级联反应,减弱了细胞生长、肿瘤增殖和细胞迁移。
