脂滴(Lipid droplets, LDs)主要由磷脂单分子层以及中性脂肪(甘油三酯和胆固醇等)疏水核心组成,并广泛存在于动物、植物和细菌中。研究显示脂滴在调节脂质代谢、自噬、细胞分化和细胞衰老等方面都发挥重要作用。
红色荧光脂滴检测试剂盒(Red Fluorometric Lipid Droplet Assay Kit)是一种通过红色荧光探针尼罗红(Nile Red)对细胞内脂滴进行检测的试剂盒。尼罗红是一种可以标记脂滴的荧光探针,其为细胞穿透性,因此可以用于活细胞和固定细胞染色。尼罗红在水或其他极性溶剂中几乎无荧光,一旦和甘油三酯等中性脂肪结合即可发出强烈的荧光。其结合中性脂肪后最大激发波长为552 nm,最大发射波长为636 nm,此时为红色荧光;也可以将激发波长设置在450-500 nm,发射波长设置为大于528 nm,此时为绿色荧光。但是尼罗红的红色荧光通常比绿色荧光明亮。由于尼罗红在红色荧光时也可以染细胞膜,在绿色荧光时只染脂滴,如果特异性检测脂滴,也可以检测其绿色荧光。
油红O也常用于脂滴检测,但油红O染色存在操作复杂、费时、准确性不高、只适用于固定细胞等问题,本试剂盒使用的尼罗红探针则没有这些问题,具有操作简单,灵敏度高等优点。但尼罗红因为发射光谱范围广,不太适合用于多重荧光染色。若需要多重荧光染色,可以考虑使用BODIPY系列探针的试剂盒。
