In vitro evaluation of cell viability, calreticulin exposure, and HMGB1 and ATP release in NCI-H929 multiple myeloma cells: NCI-H929 cells were treated with GSK2857916 (0.039–2.5 μg/ml, 48 h); mitoxantrone (1 μM) was the positive control (ICD inducer). 2. In vitro activation of immature DCs by GSK2857916-treated NCI-H929 cells: GSK2857916-treated NCI-H929 cells (0.1–10 μg/mL, 48 h) were co-cultured with monocyte-derived, day 7 immature DCs at a 1:1 ratio (7.5×10⁴ cells/well, 96-well plate) for 24 h; DC surface markers were analyzed by flow cytometry. 3. In vitro growth death assays: NCI-H929/EL4-hBCMA cells underwent 72-h treatment with GSK2857916 (0-10,000 μg/mL). |
