背景

XTT (sodium salt), a second-generation water-soluble variant of the tetrazolium salt 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), measures cell proliferation based on the metabolic activity of the cells [1].

XTT, a yellow water-soluble substrate, can be metabolically reduced by mitochondrial dehydrogenase in viable cells to an orange water-soluble formazan product. Thus, XTT has been widely used for evaluation of cell viability [2].

In human larynx cancer cell line (HEp-2), exposed to cyclophosphamide (0.005 ~ 100 µM) for 8, 12 and 24 hrs, XTT was used for determination of the IC50 value cyclophosphamide. XTT assay was sensitive enough to detect early cytotoxicity starting after incubation of HEp-2 cells with cyclophosphamide for 8 hrs, better than MTT assay which revealed cytotoxicity due to cyclophosphamide exposure after 12 hrs [3]. In addition to its application in quantitative colorimetric assay, XTT exhibited anti-HIV-1 activity by forming XTT formazan. In MT-2 cells, pre-incubation of XTT (200 μg/ml) plus phenazine methosulfate (PMS) resulted in inhibition of HIV-1-mediated cell-to-cell fusion [2].
