肿瘤坏死因子α (Tumor Necrosis Factor alpha, TNF-a)是一种主要由巨噬细胞和其它一些类型细胞如CD4+淋巴细胞、NK细胞等产生的促炎细胞因子。TNF-α生物学作用有杀伤肿瘤、促进炎症反应、抗病毒、发热反应与免疫调节等。TNF-α在其他多种细胞中也有较低的表达,如成纤维细胞、平滑肌细胞和肿瘤细胞。研究表明TNF-α不仅对肿瘤细胞具有细胞毒性,还可以诱导产生多种的白细胞介素和干扰素,这些细胞因子的网络活性可以相互协同,形成一系列连锁放大反应,从而加重炎症反应,最终导致机体的大范围损伤,比如感染性休克、多器官损伤等危重症的发生。TNF-α还具有对骨组织的吸收作用而造成对骨的破坏,目前己知的与TNF-α相关联的疾病包括:艾滋病、贫血、肿瘤、出血性休克、器官移植的排斥反应、结核病、白血病、糖尿病、类风湿性关节炎等。近些年来,TNF-α.也是单克隆抗体和蛋白酶抑制剂等相关领域极具重要意义的靶点分子。
TNF-α以分泌的可溶性形式和膜锚定形式出现,两者均具有生物活性。天然存在的TNF-α形式是糖基化的,但非糖基化的重组TNF-α具有相当的生物活性。据报道,TNF-α的生物活性天然形式是三聚体。人与小鼠TNF-α在氨基酸水平.上和交叉反应性上表现出约79%的同源性。
肿瘤细胞株对TNF-α的敏感性有很大的差异,用放线菌素D、丝裂酶素C、放线菌酮等处理肿瘤细胞,可明显增强TNF-α杀伤肿瘤细胞活性。
