Fluo-3 (potassium salt)是Fluo-3的钾盐形式,是一种Ca2+指示剂,可用于检测细胞中的游离Ca2+。Fluo-3钾盐不能穿透活细胞膜,但可以通过显微注射(microinjection)或刮擦法(scrape)装载到细胞中。
细胞内的Fluo-3以游离配体形式存在时几乎是非荧光性的,但是当它与细胞内钙离子结合后可以产生较强的荧光,荧光会增加60至80倍,最大激发波长为 506nm,最大发射波长为 526nm。可以使用激光共聚焦显微镜或流式细胞仪检测细胞内钙离子浓度的变化[1,2]。
与许多其他Ca2+指示剂(如Fluo-4,Calcium Green-1,Calcium Orange,Oregon Green 488 BAPTA-1和Fura-Red)相比,Fluo-3被证明是最适用的Ca2+指示剂,但是更容易发生光漂白[3]。
