Pyridostatin是合成的G-四联体小分子稳定剂[1]。
G-四联体是一种DNA二级结构,通常存在于染色体或端粒的末端。由于G-四联体也存在于一系列原癌基因(包括c-kit、K-ras和Bcl-2)的启动子中,因而被认为参与基因复制和转录的调节过程。此外,G-四联体可以影响端粒延伸、复制和封盖。基于上述发现,许多与G-四联体相互作用的小分子已被设计并合成,用于验证G-四联体的存在与作用,或被研发作为选择性抗癌药物。据报道,一些与G-四联体相互作用的小分子可以引起端粒逐渐缩短,从而促进DNA损伤应答,最终使细胞周期停滞。在上述分子中,Pyridostatin是合成的G-四联体小分子稳定剂,可以适应G-四联的动态与多样化结构。Pyridostatin与端粒相关蛋白竞争结合,诱导端粒功能障碍[1,2]。
在FRET熔解实验中,利用人类端粒G-四联体形成序列和双链DNA,Pyridostatin在1 μM的浓度下对G-四联体显示出最强的稳定效果,同时不影响双链DNA。在癌细胞系(HeLa、U2OS和HT1080)和正常细胞系(WI-38)中,给予Pyridostatin 72小时后显著抑制细胞生长,其IC50值范围为0.89 ~ 10 μM。Pyridostatin对HT1080细胞的选择性比对正常细胞的高18倍[1,3]。
