7-Aminoactinomycin D(7-AAD)是一种DNA荧光染料,与DNA结合后会发出强烈的荧光。7-AAD不具有细胞膜渗透性,无法进入活细胞和早期凋亡细胞中进行染色,但是可以对丧失膜完整性的受损细胞、坏死细胞和晚期凋亡细胞进行染色。7-AAD也可以对已固定并通透的细胞进行染色。7-AAD可用于多色荧光显微镜和流式细胞仪。
由于7-AAD选择性地插入富含GC的DNA区域,从而在多线染色体(polytene chromosome)和染色质中产生独特的谱带模式,这使得该染料可用于染色体谱带研究。
7-AAD / DNA复合物可以用氩离子激光在488 nm处激发,最大发射峰为647 nm。7-AAD荧光特性与PI相似,可被488nm氩离子激光激发,但其发射光谱较PI窄,且发射波长更长,通常用作碘化丙啶(PI)的替代品,以区分和排除流式细胞仪分析中的非活性细胞[1]。与FL2通道的PI检测相比,7-AAD可以在FL3通道检测。这使得7-AAD可以与不会在FL3通道检测的荧光染料一起使用,如PE(藻红蛋白),FITC(异硫氰酸荧光素)和APC(异藻蓝蛋白)等。Annexin V / 7-AAD染色可以区分早期凋亡与晚期凋亡和坏死[2]。
