YC-1对缺氧诱导因子-1(HIF-1)转录活性的IC50值为1.2 μM。
YC-1是一种新型的抗癌药物,最初被开发作为一种HIF-1α抑制剂。缺氧诱导因子-1α(HIF-1α)是一种碱性螺旋-环-螺旋转录因子,调控VEGF和其它基因表达,调节低氧条件下的肿瘤细胞存活、生长和转移。
体外:YC-1激活可溶性鸟苷酸环化酶,从而抑制血小板聚集与血管收缩,最初被研发用于治疗循环障碍。在低氧条件下的肝癌细胞中,YC-1在转录后水平上完全抑制HIF-1α表达,从而阻断HIF-1的转录因子活性。结果表明,YC-1的上述作用可能与氧感受通路相关,而与可溶性鸟苷酸环化酶活化无关[1]。
体内:与赋形剂组小鼠的肿瘤相比,YC-1治疗组的肿瘤具有显著减小的体积以及明显较少的血管。此外,在YC-1治疗组的肿瘤中,无论肿瘤类型,HIF-1α以及HIF-1诱导性基因的表达水平较低[1]。
