Vortioxetine (Lu AA21004) HBr

Vortioxetine (Lu AA21004) HBr（vortioxetine）是一种抗抑郁药 [1][2]和血清素（5-HT）受体调节剂[1][3]，对人类（h）5-HT3A受体、h5-HT7受体、大鼠（r）5-HT7受体、h5-HT1B受体、h5-HT1A受体和h5-HT转运子（hSERT）的Ki值分别为3.7 nM、19 nM、200 nM、33 nM、15 nM和1.6 nM，对h5-HT1B受体的EC50值为460 nM[3]。

5-HT是一种神经调节性神经递质，在大脑中十分重要[4]。

在稳定表达h5-HT1B的HELA细胞中，Lu AA21004具有部分激动剂响应，EC50值为460±110 nM，IA为22%。在表达r5-HT7受体的多克隆HEK-293细胞系中，Lu AA21004结合r5-HT7受体，Ki值为200±27 nM，对r5-HT7受体是一种功能性拮抗剂，IC50值为2080±18 nM[3]。

在大鼠中，皮下（sc）使用Lu AA21004对rSERT和r5-HT1B受体的ED50值分别为0.4 mg/kg和3.2 mg/kg。Lu AA21004作为5-HT3受体的拮抗剂，ED50值为0.11 mg/kg sc[3]。在大鼠中，使用剂量为2.5和5 mg/kg的vortioxetine，发现新物质的平均检出时间分别为29和33 s。
