UNC3866是一种强效的CBX4和CBX7 chromodomains拮抗剂,Kd值均为约100 nM,选择性是其他七种CBX和CDY chromodomains的6至18倍。UNC3866拮抗chromodomains的Polycomb CBX和CDY家族的甲基赖氨酸(KME)阅读功能。[1]
UNC3866是已报道的最强效的CBX7配体,Kd为 97±2.4 nM。通过AlphaScreen(IC50 = 66±1.2 nM)检测,UNC3866拮抗CBX7-H3的相互作用。UNC3866对CBX2、CBX4、CBX6和CBX8的亲和性与每个chromodomain和CBX7的序列同一性百分比紧密相关联。UNC3866对最类似于CBX7的CBX4同等强效,对CBX4和CBX7的选择性分别为CBX2、CBX6和CBX8的18、6和12倍以上。UNC3866对CBX4和CBX7的选择性超过CDY1的65倍,超过CDYL1b和CDYL2的选择性9倍。UNC3866每个骨架酰胺参与和CBX7骨架结合的至少一个氢键,而UNC3866的N-末端叔丁基苯甲酰帽主要连接D50、R52和L53的侧链。X射线晶体学显示,UNC3866和CBX chromodomains的相互作用密切模拟甲基化H3尾部的相互作用。UNC3866抑制PC3细胞增殖,与CBX7过表达以赋予生长优势的已知能力相一致。[1]
