UNC2881是Mer酪氨酸激酶的选择性抑制剂[1].
Mer是TAM (Tyro3\Axl和Mer)受体酪氨酸激酶(RTK)亚家族的一员,生长停滞特异性蛋白6(Gas6)是其内源性配体.Mer在血小板凝集和巨噬细胞活性中发挥重要作用.
UNC2881是新型的Mer酪氨酸激酶特异性抑制剂.在激酶抑制试验中,UNC2881(430 nM)抑制超过50%的MER\FGFR1\KDR\EGFR\ITK和TRKA.在697 B-ALL细胞中,UNC2881抑制Mer磷酸化,IC50值为22 nM.UNC2881能够抑制Mer胞内域和上皮生长因子受体(EGFR)胞外域组成的嵌合蛋白的配体依赖性磷酸化.在人血小板富集的血浆中,UNC2881抑制超过25%的纤维状I型马胶原蛋白诱导的血小板凝集.UNC2881也能抑制ATP释放,其是血小板活化的标志[1].
在小鼠中,UNC2881具有14%的口服生物利用度,以及高的全身清除率.UNC2881在体外或短期体内研究中是有效的[1].
