TCS HDAC6 20b是组蛋白去乙酰化酶6(HDAC6)的有效选择性抑制剂[1]。
去乙酰化酶(HADC)可以除去组蛋白中ε-N-乙酰赖氨酸氨基酸上的乙酰基,使组蛋白更紧密地包裹住DNA,阻止转录。HADC6的突变与阿尔茨海默病相关。
在HCT116细胞中,TCS HDAC6 20b以剂量依赖性方式增加ɑ微管蛋白的乙酰化,而乙酰化组蛋白H4没有明显增加,这表明,TCS HDAC6 20b选择性抑制HDAC6[1]。使用剂量为5 μM的TCS HDAC6 20b和剂量为0.03 μM紫杉醇(PTX)处理人结肠癌HCT116细胞,抑制了约50%的细胞生长,这表明,TCS HDAC6 20b可以与PTX一起使用,作为潜在的抗癌药物。在雌激素受体ɑ(ERɑ)阳性乳腺癌MCF-7细胞中,使用1 nM 的17β雌二醇(E2),细胞的生长提高40%,TCS HDAC6 20b显著阻断这个过程[2]。
