TBA354是一种含吡啶的联芳基化合物,用于对抗结核分枝杆菌.在10个检测的结核分枝杆菌菌株中,TBA-354的MIC值范围为小于0.02 μM -0.36 μM.对金黄色葡萄球菌\大肠杆菌和白色念珠菌的MIC值均大于50 μM,这表明TBA-354是一种窄谱剂.在Caco-2细胞的双向渗透实验中,1 μM和10 μM TBA-354具有高渗透性.TBA-354与肝微粒体一起孵育时,其代谢稳定或中度代谢.在HLM中,TBA-354(30 μM)较弱地抑制CYP3A4催化的睾酮6-羟化(40%)[1].
在低剂量气溶胶感染模型中,给予TBA-354,每天100 mg/kg,每周5天,连续3周,具有显著杀菌活性并降低肺CFU(约1 log10).在慢性结核病的小鼠模型中,TBA-354呈剂量与时间依赖性地杀死结核分枝杆菌,并表现出显著杀菌活性.TBA-354具有较高的生物利用度和较长的半衰期[1].在结核病小鼠模型中,TBA-354显著增加Bedaquiline的杀菌活性[2].
