SR9011是一种REV-ERB-α/β激动剂。
REV-ERB核受体在昼夜节律的反馈调节中具有关键作用。Bmal1和Clock都是REV-ERB的直接靶基因,并且REV-ERB-α的丧失改变昼夜节律行为。
体外:在HEK293细胞中,SR9011剂量依赖性地增加REV-ERB依赖性阻遏物活性。此外,使用全长REV-ERB-a以及由Bmal1启动子驱动的荧光素酶报告基因进行共转染试验,SR9011强有效地抑制转录。SR9011还以REV-ERB-a/b依赖性方式抑制HepG2细胞中BMAL1信使RNA的表达。此外,SR9011抑制SCNclock的活性,对昼夜节律具有可逆抑制[1]。
体内:在恒定黑暗条件下的小鼠中观察到,跑轮行为的SR9011依赖性减少是剂量依赖性的,并且效力(半数最大有效剂量为56 mg/kg)类似于SR9011介导的对REV-ERB应答基因Srebf1的抑制效力。此外,Tau不受SR9011治疗的影响,给药后正常节律的恢复与从SCN外植体中移除药物后观察到的效果相似[1]。
临床试验:到目前为止,SR9011仍处于临床前研究阶段。
