SR1078是一种选择性的RORα和RORγ激动剂,通过诱导构象变化来激活RORα和RORγ介导的转录[1].
SR1078作为一种激动剂选择性靶向RORα和RORγ,而非LXR和FXR[1].结果表明,SR1078能够刺激ROR活性和随后ROR介导的mRNA表达.当用SR1078处理HepG2细胞24小时后,ROR介导的报告基因FG21的mRNA水平提升了3倍,而另一个报告基因G6Pase提升了2倍[1].
在小鼠模型中,腹腔注射10 mg/kg 的SR1078一小时导致血浆浓度为3.6 μM,八小时后该浓度仍维持在800 nM.腹腔注射10 mg/kg 的SR1078两小时可显著提高报告基因G6Pase和FG21的信使RNA水平[1].
