Solamargine是一种有毒的化合物,存在于茄科植物,如土豆、西红柿和茄子中[1]。 从茄属(Solanum nigrum)真菌内生菌黄曲霉(Aspergillus flavus)中可分离得到Solamargine[2]。
体外实验:Solamargine通过下调MMP-2和MMP-9的表达和活性,显著抑制HepG2细胞的迁移和侵袭。Solamargine以浓度依赖性方式降低HepG2细胞活力。在HepG2细胞中,solamargine(7.5 μM)降低小于20%的细胞活力。最高剂量Solamargine分别减少超过70%和72%的细胞迁移和侵袭。Solamargine还降低MMP-2和MMP-9蛋白的表达和功能[3]。在肺癌细胞中,Solamargine通过p38 MAPK介导的Stat3磷酸化和蛋白表达的抑制,进而抑制增殖并诱导细胞凋亡,随后诱导Stat3下游效应物p21。在非小细胞肺癌(NSCLC)细胞中,Solamargine以时间和剂量依赖的方式抑制生长并诱导细胞凋亡。Solamargine还以时间依赖性方式增加p38促分裂原活化蛋白激酶(p38 MAPK)的磷酸化[4]。在SMMC-7721细胞中,solamargine通过活化caspase-3和调节细胞周期进程来诱导细胞凋亡和抑制肝癌细胞增殖,进而发挥潜在的抗癌活性[5]。在WM115和WM239细胞中,Solamargine快速、有效和选择性抑制WM239和WM115细胞的生长。Solamargine对垂直生长期黑色素瘤细胞具有较高效力[6]。
