Phosbind是一种新型的磷酸盐结合标签和功能分子,在中性pH(生理pH)条件下可以特异性结合磷酸离子。Phosbind是一种双环金属络合物(1,3-bis[bis(pyridin-2-ylmethyl) amino]propan-2-olato dizinc(II) complex),是一种选择性的磷酸结合标签,在中性pH的水溶液中,其对苯基磷酸酯二价阴离子(PH-OPO32-)的Kd值为25 nM* (*:在Zn2+的情况下)。
Phosbind Acrylamide提供了一种特异性的电泳过程(锰 (II)- Phosbind SDS-PAGE),可同时对磷酸化蛋白及其对应的非磷酸化形式进行分析。
此产品最适合做的蛋白大小在30 KDa-130 KDa之间

4. 在制备SDS-PAGE凝胶时只需在丙烯酰胺溶液中简单加入Phosbind Acrylamide和MnCl2溶液*即可。
(*:由于1个Phosbind可结合2个锰离子(Mn2+),在凝胶中加入5.0 mM Phosbind Acrylamide水溶液和10 mM MnCl2溶液。)
(1) 染色
• 适于激酶实验。
• 与常规SDS-PAGE几乎相同的机制。
(2) Western Blotting
• 不需要准备针对磷酸化蛋白的抗体。
• 可用于内源性蛋白的磷酸化分析。
• 不需要放射性同位素标记。
