Jingzhaotoxin III是一种选择性的Nav1.5通道阻断剂,IC50值为0.348 μM[1]。Nav1.5是一种电压门控钠通道,主要在心肌细胞中表达,对心脏的电生理活动起着重要的调控作用。异常的Nav1.5通道活性与多种心脏疾病,如心律失常、心肌梗死等有关。
在细胞水平上,Jingzhaotoxin III能有效阻断Nav1.5通道的活性,从而影响心肌细胞的电生理活动。研究发现,Jingzhaotoxin III对Nav1.5通道的阻断作用具有剂量依赖性,当浓度达到1.03 μM时,可以完全阻断Nav1.5通道的活性[2]。此外,Jingzhaotoxin III对Nav1.5通道的阻断作用具有快速性和可逆性,这使得其在临床应用中具有潜在的优势。
在动物模型中,Jingzhaotoxin III的应用可以有效改善心律失常的症状。在一项对小鼠心律失常模型的研究中,通过给予Jingzhaotoxin III治疗,可以显著降低小鼠的心律失常发作频率和严重程度[2]。这些研究结果表明,Jingzhaotoxin III作为Nav1.5通道阻断剂,具有很好的抗心律失常效果,为其在心律失常等心脏疾病的治疗中的应用提供了理论依据。
