5-溴脱氧尿苷(5-BrdU)是胸苷类似物,在细胞处于S期时掺入DNA,并能被单抗或多抗检测出.
体外实验:在粉蓝烟草髓外植体生长的去分化早期研究5-BrdU对增殖的作用.只有在培养的第一个72小时期间给予5-BrdU才能完全抑制,此抑制作用能被同时加入脱氧胞苷或胸苷所逆转[1].
体内试验:年轻成年后代小鼠中使用单抗和过氧化物酶-抗过氧化物酶方法对5-BrdU染色.免疫反应核分布与之前[3H]胸苷的数据相似.不同胚胎期的标记核模式遵循皮层和海马神经发生所表现的时空时间梯度.这些数据表明BrdU能够用于反应神经元出现时期[2].
临床试验:目前尚无临床数据.

应用 |
在烟碱髓组织中,BrdU抑制了髓外植体的生长,而胸苷完全逆转了抑制作用。观察到的抑制作用可归因于BrdU掺入DNA。 |

应用 |
在怀孕的NMRI小鼠中,在胚胎期E11和E17之间注射单次脉冲的5-bromodeoxyuridine (BrdU)以标记有丝分裂后神经母细胞。BrdU是Thy的类似物,能够插入经历S期的细胞的DNA中,并且可以通过多克隆或单克隆抗体检测。在BrdU脉冲后,在成人CNS中检测到免疫反应性核。BrdU可用于映射神经元的出生日期。 |
