多巴胺受体是一类G蛋白偶联受体,在中枢神经系统中有突出地位.多巴胺受体参与多种神经过程.因此,多巴胺受体是常见的神经药物靶点.抗精神病药经常是多巴胺受体拮抗剂,而典型的神经兴奋剂则通常是多巴胺受体的间接激动剂.Chlorpromazine是一种多巴胺拮抗剂.
在体外:chlorpromazine的抗精神病活性与其拮抗多巴胺受体的功能有关,其具有吩噻嗪环结构,可与多巴胺受体相互作用.此外,chlorpromazine可抑制[3H]螺哌隆的结合,其抑制曲线与单一位点结合的类型相一致[1].
在体内:大鼠每天使用chlorpromazine持续21日可导致僵住症\对僵住症的耐受和PCP挑战带来的自主活动敏化.结果表明每日chlorpromazine治疗可诱导DA/NMDA受体对PCP挑战带来的总的自主活动敏感[2].
临床试验:Chlorpromazine在临床上是一种传统的抗精神病药,自从1954年被FDA批准后一直用于处理精神分裂.

10 ~ 100 μM的Chlorpromazine HCl呈剂量依赖性地降低mIPSC幅度。此外,当浓度≥ 30 μM时,Chlorpromazine HCl呈剂量依赖性地显著加快mIPSC衰减时间。然而,对照组与Chlorpromazine HCl治疗组的10 ~ 90%上升时间无显著差异。 |

在缺氧大鼠模型中,Chlorpromazine HCl减少脑组织突触传递的不可逆缺失。Chlorpromazine HCl还可以减缓Ca2+流入神经元,从而显著延迟缺氧诱导扩散抑制的发生。 |
