StemRegenin1(SR1),嘌呤衍生物,是一种有效的芳香烃受体(AhR)小分子抑制剂,在CD34+细胞中有效拮抗AhR,IC50值为127 nM。SR1可以促进CD34+细胞的体外扩增。最近的研究表明,在造血干细胞(HSC)中,SR1增加了表达CD34的细胞和保留嫁接免疫缺陷小鼠能力的细胞,分别增加了50倍和17倍。SR1也可诱导pDCs、BDCA1+和BDCA3+ mDCs的体外分化,及所有这些树突状细胞(DC)亚群的高细胞数量的产生。

该化合物在DMSO中的溶解度大于10 mM,若配制更高浓度的溶液,一般步骤如下:请将试管置于37℃加热10分钟和/或将其置于超声波浴中震荡一段时间。原液于-20℃可放置数月。 |

实验结果 |
用1 μM SR1或0.01% DMSO(对照)培养CD34+ UCB细胞,持续3周。在第7、14和21天测定总细胞数,并通过流式细胞术在每个时间点评价pDC的频率。在3周的培养期间,总成核细胞不断增加。时间点的pDC分化分析表明,在第14天达到pDCs的最大频率。在第21天,pDCs的绝对数目最高。 |
