Tubacin是HDAC6有效的、选择性、可逆的、细胞通透性的抑制剂,IC50值4 nM[1]。
组蛋白去乙酰酶(HDACs)可分为4类,其中类I、II和IV是核锌依赖性酶,类III是烟酰胺腺嘌呤二核苷酸(NAD+)依赖性酶。HDACs催化N-乙酰半胱氨酸残基脱乙酰,在很多生物反应包括基因表达和细胞周期中发挥重要作用。在哺乳动物细胞中,通过抑制α微管蛋白脱乙酰基,Tubacin可以抑制特定基因的表达,达到无组蛋白乙酰化水平的抗肿瘤效应。在蛋白质降解变性的过程中使用HDAC6的选择性抑制剂处理,表明Tubacin可能具有抗肿瘤转移和抗血管生成的疗效[1,2]。
Tubacin是HDAC6有效的抑制剂(IC50值4 nM),对HDAC6的抑制强度几乎是HDAC1的350倍。在培养的A549细胞中,tubacin(10 μM)诱导α-微管蛋白的乙酰化水平增加3倍,EC50值2.5 μM。用0.5 - 2.5 mM不同浓度的Tubacin治疗急性成淋巴细胞性白血病(Acute lymphoblastic leukemia,ALL)和正常细胞,并设置对照。结果表明Tubacin剂量依赖性地抑制ALL细胞的生长,IC50值1.2 - 2 mM。此外,ALL细胞相比其他正常细胞对Tubacin更敏感[1,2,3]。
Tuacin在体内也对ALL细胞的生长表现出抑制活性。将前B ALL细胞注入老鼠,实验组老鼠相比于对照组寿命延长。此外,Tubacin处理转染了τ的HEK细胞(此细胞在T231显著降低了τ磷酸化),也表明Tubacin在阿尔茨海默病(AD)方面可能发挥了重要作用[3,4]。
