WR-1065是一种氨磷汀(Ethyol)的去磷酸化代谢产物,能防止辐射直接和延迟的影响。
WR-1065可防止齐多夫定(AZT)诱发的基因毒性。淋巴母细胞系MOLT-3细胞暴露在0/10 μM的AZT中96 h。在开始的24 h,加入0/5 μM的WR-1065,76 h时加入Cyt B,AZT处理过的MOLT-3细胞的活力未发生变化。[1]此外,RKO36细胞系(携带GFP-pCMV-EGFP2Xho 的RKO人类结肠癌衍生系)在放射前迅速以终浓度为4 mM(EC50)的WR-1065处理,可体现对电离辐射和迟发的基因组不稳定性引发的细胞染色体损伤和死亡的保护作用。但是40 μM的WR-1065在已被辐照的RKO36细胞中不体现即时的射线防护作用。[2] WR-1065主要通过抑制同源重组途径体现其作为辐射防护剂的功能。在SPD8中国仓鼠细胞系中,4 mM的WR-1065处理30 min和10 μM处理24 h可显著减少0.2 mM羟基脲处理24 h或100 nM喜树碱处理1 h引起的同源重组。但是相比同源重组高效的细胞AA8/CXR3,WR-1065并未在同源充足缺陷的irs 1SF细胞中显示辐射防护功能(P<0.05)。[3]
使用PLGA(聚乙丙交酯)为聚合物基质的喷雾干燥技术可将WR-1065制成纳米粒子。以小鼠口服含有21.7(w/w WR-1065)的500 mg/kg的WR-1065/PLGA纳米粒子的方式来确定其辐射防护功能。相对于未经处理的对照组小鼠,WR-1065PLGA纳米粒子处理的小鼠具有显著更高的30天的存活、较少的骨髓抑制和肠损伤,显示其突出的辐射防护效果。[4]
