X-Neu5Ac是一种用于细菌表达系统中神经氨酸酶活性显色试验的新底物。

体外:X-Neu5Ac被神经氨酸酶水解释放出卤化产物,经历迅速的有氧氧化以形成深蓝色色素。初步动力学研究表明,X-Neu5Ac是一种良好的和稳定的神经氨酸酶底物。此外,X-Neu5Ac也被突变酶水解[1]。
体内:用X-Neu5Ac在pH 7.3下培养急性脑切片,以显现大鼠脑膜表面的细胞外唾液酸酶活性。1小时后,大鼠脑髓鞘丰富区域表现出强烈的荧光。尽管大脑海马表现出微弱荧光,海马苔藓纤维终端表现比较强烈的荧光。另外,X-Neu5Ac引起的荧光强度与唾液酸酶活性相关。因此,X-Neu5Ac染色是唾液酸酶特异性的,可用于唾液酸酶活性的定量分析[1]。
