体外:100 μM 的NNC 55-0396对高电压钙通道没有影响,但在HEK293细胞中抑制T型通道,效力比得上mibefradil(IC50为 6.8对10.1 μM)[1]。
体内:在水平丝试验中,NNC 55-0396比mibefradil具有更好的耐受性。小鼠给药NNC 55-0396,随后给药harmaline或媒介物,以评估倒置线栅测试中与harmaline的潜在相互作用。NNC 55-0396改善harmaline诱导的试验缺陷。在GABAA α1-null模型中,12.5 mg/kg 的NNC 55-0396在20-100分钟内抑制半数harmaline诱导的震颤,而相同剂量的mibefradil没有显著影响震颤。对比mibefradil,NNC 55-0396具有良好的耐受性,并且抑制震颤,且具有较低的细胞色素P450抑制作用[1]。
