LDN193189是骨形态发生蛋白(BMP)I型受体转录活性的选择性抑制剂,抑制活化素受体样激酶2(ALK2)和ALK3,相应IC50值为5 nM和30 nM[1]。
在C2C12细胞中,LDN193189可以抑制BMP诱导的Smad信号(Smad1/5/8)和非Smad信号磷酸化,包括p38和Akt[2]。
在支气管上皮(Beas2B)细胞和C57BL/6小鼠中,LDN193189对BMP的药理抑制作用可以防止BMP2诱导的E-钙粘蛋白下调。在细胞水平上,LDN193189也能抑制BMP诱导的上皮通透性减少[3]。

在出生后第7天(P7)的Ad.Cre条件性CaALK2转基因小鼠中,腹腔注射LDN-193189(3 mg/kg)导致左胫骨周围的轻度钙化,引起P13上产生可见的腓骨,并且防止P15处的放射损伤,不引起体重减轻或发育迟缓、自发性骨折、骨密度降低或行为异常等现象。在PCa-118b肿瘤小鼠中,LDN-193189减轻肿瘤生长并减少肿瘤中的骨形成。 |
