AEBSF是一种不可逆的、广谱性丝氨酸蛋白酶抑制剂[1]。
AEBSF是蛋白酶的共价结合抑制剂,包括胰蛋白酶、胰凝乳蛋白酶、纤溶酶和凝血酶。AEBSF可抑制各种细胞系中Aβ的生成。在转染APP695(K695sw)的K293细胞中,AEBSF剂量依赖性地减少Aβ,IC50值约为1 mM。在转染野生型APP695的HS695和SKN695细胞中,AEBSF也具有抑制效应,IC50值约为300 μM。AEBSF也可以增加α-切除,抑制β-裂解。此外,作为蛋白酶的抑制剂,AEBSF阻止单核细胞源性的巨噬细胞裂解白血病细胞。巨噬细胞与150 μM的AEBSF孵育6小时后几乎导致最大程度的抑制[1,2]。

实验结果 |
据发现,在各种细胞系中,AEBSF抑制Aβ生成。在转染了βAPP695(K695sw)的K293细胞中,AEBSF呈剂量依赖性地减少Aβ,其IC50值约为1 mM。在转染了野生型APP695的HS695和SKN695细胞中,AEBSF显示出抑制作用,其IC50值约为300 μM。此外,AEBSF增加α裂解并抑制β裂解。 |
