Quercetin是重要的膳食类黄酮,存在于蔬菜、水果、种子、坚果、茶和红酒中。该物质具有多种与肿瘤预防相关的生物学功能。Quercetin是PI3K、NF-κB和其他参与胞内信号的激酶的强抑制剂。该物质也可轻度抑制Akt1/2,并对PKC、p38和ERK1/2有微弱效果[1]。
多个报道证明,Quercetin在癌细胞中具有抗炎、抗氧化和促凋亡功能,可通过多种机制抑制肿瘤进程。Quercetin可提高胞质Ca2+水平,破坏线粒体膜电位(MMP),反过来促进胞质中细胞色素c释放,从而激活多种caspases,如caspase-3、-8和-9,进而通过线粒体途径诱发细胞凋亡[2]。Quercetin可诱导细胞周期停滞和细胞凋亡,这与p53的功能有关。Quercetin可促进p53磷酸化,从而在mRNA和蛋白质水平稳定p53[3]。然而,多个研究表明,含有受损p53的细胞对Quercetin介导的细胞毒性和凋亡更为敏感[4]。
体内研究表明,在致癌作用发生前服用Quercetin可显著降低小鼠模型中多种化学制剂诱导的肿瘤负荷。包括苯并(a)芘诱导的肺肿瘤负荷、大鼠结肠中氧化偶氮甲烷诱导的癌前病变和N-亚硝胺诱导的肝癌等[5]。
