Myristoylated PKI (14-22) amide是一种有效的cAMP依赖性蛋白激酶(PKA)抑制剂,阻断脊柱施用8-bromo-cAMP所产生的痛觉过敏[1]。
PKAs是真核生物中cAMP信号的主要介体,在几个生物学过程,如基因表达、细胞凋亡、组织分化和细胞增殖中发挥重要作用。PKAs通过对蛋白底物的丝氨酸/苏氨酸残基进行磷酸化而发挥这些功能[2]。
PKAs在嗜中性粒细胞的吞噬作用中起关键作用。CAMP/PKAs在受体介导的吞噬作用中调节F-肌动蛋白的重组,这是由IgG-FcR的相互作用而触发的。Myristoylated PKI 14-22 amide以剂量依赖的方式减少IgG介导的吞噬反应。在浓度高于10 μM时,PKI 14-22 amide可以抑制中性粒细胞粘附,这使得吞噬的测量难以执行[1]。
由于PKA在哺乳动物细胞中活性的失调与几种类型癌症的发病机理相关,PKI (14–22) amide可用作潜在的疗法,用于治疗这些癌症和许多与PKAs相关的其它疾病[1,2]。
