PETCM是Caspase-3的激活剂[1]。
Caspase-3属于半胱氨酸天冬氨酸蛋白酶家族,并与caspase-8和caspase-9相互作用。Caspases的顺序激活在细胞凋亡中起重要作用。
在HeLa S-100细胞胞质溶胶中,PETCM以剂量依赖性方式活化caspase-3,诱导凋亡体的形成。0.2 mM的PETCM引起Apaf-1转移到约1百万道尔顿的大小,意味着凋亡体的形成。此外,PETCM需要dATP发挥功能。PETCM对caspase-3的活化作用独立于PHAP蛋白质。当PHAP蛋白的激活活性被抑制时,PETCM可以逆转这种抑制。当Q30(Apaf-1/procaspase-9)与细胞色素c和10 μM的dATP加入时,重组PHAPI激活caspase-3。重组ProT抑制这种活性。同时,PETCM逆转ProT的抑制作用。PHAPI不影响凋亡体的形成,活化caspase-9和Apaf-1,这些与凋亡体相关。ProT有效地阻止凋亡体的形成,而PETCM缓解这种影响[1]。
