Pam3CSK4是Toll样受体1/2(TLR1/2)的激动剂[1]。
Toll样受体(TLR)在先天免疫系统中发挥重要作用。TLR主要表达于前哨细胞中,识别由微生物衍生的分子,然后激活免疫细胞应答。
Pam3CSK4是TLR1/2的激动剂。Pam3CSK4以Syk和src依赖性方式增加PLCγ2和接头蛋白LAT的酪氨酸磷酸化。此外,Pam3CSK4诱发血小板活化[1]。
在对粉尘螨过敏原过敏的小鼠鼻炎模型中,Pam3CSK4显著改善鼻过敏症状,并降低支气管肺泡灌洗液(BALF)中总炎症细胞和嗜酸性粒细胞。此外,Pam3CSK4显著增加IFN-γ的释放,减少IL-13和总IgE的水平 [2]。Pam3CSK4诱导巨噬细胞产生NO和TNF-ɑ[3]。在用OVA激发的小鼠哮喘模型中,Pam3CSK4显著降低BALF中嗜酸性粒细胞和总炎症细胞渗透,并抑制OVA诱导的IL-4和IL-5的分泌。此外,Pam3CSK4减少IgE的水平,增加IL-12的生成[4]。
