MPEP是一种选择性的和具有全身活性的代谢型谷氨酸受体亚型5(mGlu5受体)拮抗剂,IC50值为36 nM[1].
MPEP是mGlu5受体的拮抗剂.该物质因其高选择性,被用作研究其靶标受体功能的一种药理学工具.在表达人类mGlu1b受体的L (tk-)细胞中,MPEP可完全抑制使君子酸诱导的PI水解,IC50值为36 nM.MPEP在表达mGlu1b受体的CHO-K1细胞或L (tk-) mGlu5a细胞系中没有作用.此外,100 μM的MPEP对第II和第III类mGlu受体亚型也没有作用.另外,MPEP可在海马体\纹状体和皮质切片中抑制DHPG诱导的PI水解,IC50值分别为8 nM\20.5 nM和17.9 nM.此外,微离子电渗入法施用MPEP可在体内降低DHPG诱导的刺激作用.MPEP在大鼠Vogel试验和小鼠尾悬浮试验中也具有抗焦虑或抗抑郁样效果[1,2].
