LY2811376是BACE1的非肽抑制剂。

β-分泌酶1（BACE1）是天冬氨酸蛋白酶，将淀粉样前体蛋白（APP）分解成β淀粉样肽（Aβ），Aβ在阿尔茨海默病（AD）中发挥关键作用。

在重组酶实验中，LY2811376抑制hBACE1，对较大的嵌合蛋白底物和小型合成肽的IC50值分别为249 nM和239 nM。在过表达APP的人胚胎肾细胞系中，LY2811376以浓度依赖性方式减少Aβ的分泌，EC50值为300 nM[1]。在SH-SY5Y细胞中，LY2811376减少分泌到细胞培养基中的Aβ1-40和Aβ1-42，同时增加Aβ5-40的水平[2]。

在APPV717F小鼠中，LY2811376显著降低BACE1裂解APP的产物，Aβ、sAPP和C99[1]。在18个健康人中，30 mg和90 mg剂量的 LY2811376以剂量依赖的方式增加Aβ5-40和Aβ5-X，同时减少Aβ1-34。这些结果表明，LY2811376通过BACE1非依赖性途径抑制N端截短的Aβ肽 [2]。
