Kartogenin是促进多能间充质干细胞(MSCs)分化为软骨细胞的强效诱导剂,EC50值为100 nM [1]。
MSCs是一种干细胞,能够分化成不同的细胞类型,包括软骨细胞、成骨细胞、脂肪细胞和肌细胞。
Kartogenin(KGN)以剂量依赖的方式有效诱导hMSCs分化成软骨细胞 [1]。在小鼠前骨骼间充质细胞中,KGN刺激软骨结节和滑膜关节的形成,增强软骨伸长率和刺激趾间内陷、肌腱成熟和域间压实。KGN增加TGFβ1的表达,而TGFβ1刺激软骨结节形成。此外,KGN增加Smad蛋白的磷酸化,Smad介导BMP和TGFβ信号 [2]。
在急性手术模型和慢性关节损伤模型中,kartogenin具有软骨保护作用。 Kartogenin打乱细丝蛋白A和转录因子核心结合因子β亚基(CBFβ)的相互作用,调节CBFβ-RUNX1转录,从而诱导软骨[1]。在BALB/C小鼠的真皮中,KGN诱导I型胶原蛋白的合成,活化TGF-β的Smad4/smad5信号传导途径[3]。
