Janex-1是一种ATP竞争性的JAK3特异性抑制剂,IC50值为78 μM [1]。
Janus激酶3(JAK3)是属于Janus激酶家族的酪氨酸激酶。通过共同的γ侧链,JAK3与Ⅰ型细胞因子受体相互作用,为受体提供酶活性。当配体结合到受体上,受体的构象发生变化,激活JAK3,由此引发下游信号。JAK3与肌动蛋白结合蛋白villin相互作用,随后促进细胞骨架重构和粘膜创伤修复。
在人类淋巴B细胞系中,Janex-1显著抑制JAK3的活性,但不影响其他类型的酪氨酸激酶,包括Janus家族酪氨酸激酶JAK1和JAK2、ZAP / SYK家族酪氨酸激酶SYK、TEC家族酪氨酸激酶BTK、Src家族酪氨酸激酶LYN。研究表明,Janex-1对JAK3的抑制是高度特异的[1]。Janex-1预处理的小鼠胰岛细胞显示出对细胞因子毒性的抗性,降低一氧化氮合酶(iNOS)的表达,从而影响一氧化氮(NO)的生成及随后的胰岛损伤。该作用的分子机制是,Janex-1通过抑制JAK3和其它信号转导,抑制iNOS的表达[2]。
在1型糖尿病NOD小鼠模型中,处理25周后,60%的对照组小鼠变为糖尿病。然而,注射Janex-1(100 mg/kg/day)10到25周,只有9%的小鼠成为糖尿病。1型糖尿病是由JAK3和下游信号引起的,糖尿病发病的抑制表明Janex-1可以抑制JAK3 [3]。
