Ivachtin是Caspase-3的有效抑制剂,IC50值为23 nM[1]。
Caspase-3是半胱氨酸天冬氨酸蛋白酶家族的成员,可与caspase-8和caspase-9相互作用。Caspases的顺序激活在细胞凋亡中起重要作用。
Ivachtin以非竞争性方式强效可逆的抑制caspase-3。Ivachtin是Caspase-3最有效的抑制剂,但对于剩余的其他caspases选择性较为适中。在用10 μM的星形孢菌素(staurosporin)处理的人Jurkat T细胞中,Ivachtin可在更高的水平保护细胞,这表明,Ivachtin具有良好的细胞渗透性和抗凋亡活性[1]。
