ITSA-1(ITSA1)是通过曲古抑菌素A(TSA)抑制的HDAC激活剂。
曲古抑菌素A(TSA)是一种链霉菌代谢物,在纳摩尔浓度可以特异性抑制哺乳动物组蛋白去乙酰化酶,在哺乳动物细胞中引起高乙酰化组蛋白分子积累。
体外:据先前的研究报告,在鼠胚胎干细胞中,与对照相比,TSA处理23小时抑制BrdU掺入。然而,在TSA单独处理被抑制的浓度,用ITSA1孵育的TSA预处理细胞可以掺入BrdU。此外,ITSA1处理能够将TSA阻滞的细胞群恢复到正常的细胞周期分布。用300 nM的TSA对A549细胞处理2小时明显增加乙酰化组蛋白H3的水平,而随后用50 μM 的ITSA1孵育2小时降低组蛋白乙酰化至基线水平。此外,在添加TSA之前用ITSA1预处理的细胞显示增加的乙酰化水平,这是TSA单独处理的特征。这些结果表明,ITSA1的靶点直到TSA诱导时才出现。此外,50或100μM 的ITSA1单独处理对HDAC活性无效,表明ITSA1可间接影响HDAC功能[1]。
体内:迄今为止,尚无动物体内数据报道。
临床试验:到目前为止,ITSA-1(ITSA1)仍处于临床前研究阶段。
