Iodoacetyl-LC-Biotin全名为N-iodoacetyl-N-biotinylhexylenediamine,是一种不溶于水的巯基反应性生物素化试剂,需要合适的溶剂,包括二甲基亚砜(DMSO)和二甲基甲酰胺(DMF),在加入到含水反应之前需要先将其溶解。Iodoacetyl-LC-Biotin含有一个生物素双环结构,一个1,6-二氨基己烷间隔臂,连接到生物素的戊酸侧链,臂末端的碘乙酰基提供巯基反应,Iodoacetyl-LC-Biotin末端的碘乙酰基能够与蛋白质和其他分子上的巯基反应形成不可逆键。Iodoacetyl-LC-Biotin的长间隔臂能够使修饰的分子更好地结合至亲和素或链霉亲和素探针。

应用 |
免疫球蛋白G(26.7 nmol)用1ml含0.1M磷酸钠和5Mm EDTA,pH 6的缓冲液溶解。在此溶液中,加入DTT使得其在溶液中的浓度为50mM,37℃室温孵育90min,然后冷却到室温。多余的DTT用pH 8.3,50mM Tris通过分子筛层析法去除,5mM EDTA用作洗脱液。随后对具有还原巯基的IgG进行生物素化,溶于Me2SO的30 μl浓度为4 mM的 iodoacetyl-LC-biotin溶液加入,混合物在室温下避光孵育90min。最后,残留的生物素化试剂和游离的生物素用pH 7.2,0.1mM 的PBS通过72小时透析除去。BCA 蛋白试验用来确定溶液中蛋白的浓度,与IgG共价结合的总biotin 通过链霉素结合试验进行检测。 |
