糖原合酶激酶-3β(GSK-3β)是一种脯氨酸定向的丝氨酸-苏氨酸激酶,参与神经元细胞发育\能量代谢和图式形成.抑制GSK-3β可导致β-连环蛋白积累.
IM-12是一种有效的GSK-3β抑制剂.在ReNcell VM细胞中,IM-12可显著提高27%的总β-连环蛋白,并在浓度为3 M时最大程度地提高β-连环蛋白量.在ReNcell VM细胞中,IM-12(3 M)可抑制GSK-3β,剩余27 ± 5%的活性.并且,IM-12可显著增加细胞倍增时间,增加到26.5 ± 0.9 h,并可显著抑制细胞增殖.在ST14A细胞中,IM-12可引发β-连环蛋白在核周围的积累.在转染TOPFlash的ReNcell VM细胞中,IM-12可提高TCF活性.在转染TOP和pCAGGS-S33Y(包含β-连环蛋白的载体)的细胞中,IM-12可显著增加270%的TCF活性.在ReNcell VM细胞中,IM-12可显著提高βIIItub+细胞水平到3.7 ± 0.7%,表明IM-12可增加神经元分化[1].
