GTS 21 dihydrochloride是一种新型的烟碱型乙酰胆碱受体(nAChRs)激动剂[1]。
nAChRs是神经元受体蛋白,被神经递质乙酰胆碱(ACh)的结合所激活。
在暴露于高氧(≥99% O2)的RAW 264.7细胞(巨噬细胞样细胞系)中,GTS-21以剂量依赖的方式显著增加巨噬细胞的吞噬活性,减少高氧诱导的HMGB1超乙酰化。GTS-21也抑制HMGB1的细胞质易位以及从巨噬细胞中的释放[1]。GTS-21与人α4β2 nAChR结合的效力(Ki值为20 nM)比人α7 nAChR高100倍,但比(2) - 烟碱与人α7和α4β2 nAChR结合的效力分别低2和18倍[2]。
暴露于高氧(≥99% O2)的小鼠,随后用PA挑战,腹腔注射GTS-21(4 mg/kg)后显著增加细菌清除,减少气道HMGB1的积累和急性肺损伤[1]。GTS-21刺激大鼠纹状体切片中多巴胺的释放,EC50值为10 μM。在延迟样本匹配任务中,GTS-21(32–130 nmol/kg)可提高猴子的学习能力[2]。
