GS967是心脏钠后期电流(late INa)的新型强效选择性抑制剂,在心室肌细胞中的IC50为 0.13 μM,在离体心脏中的IC50为 0.21 μM[1].
当肌细胞Na+通道开放后不能失活,钠离子内流贯穿整个动作电位(AP).导致的Na+电流(INa)被称为晚期INa.在许多病理状况下其幅度增加,如衰竭或缺血性心脏\暴露于氧化应激的心脏以及先天性长QT3综合征患者的心脏[1].
在分离的兔心室肌细胞中,GS967通过浓度和电压依赖性方式以最小的用量抑制峰INa,也降低Na+和Ca2+超载.在兔离体心脏中,GS967废除ATX-II或E-4031诱导的心动过速[1].
在麻醉兔中,GS967减少MAPD90,但并没有改变心脏传导时间,它也防止氯非铵诱导的心律失常活性和心动过速,降低局部缺血诱导的心律失常的发生率[1]
