抗逆转录病毒蛋白酶是蛋白酶抑制剂的一个亚家族,在治疗HIV/AIDS和HCV感染中具有重要作用。据报道,蛋白酶抑制剂药物通过选择性结合病毒蛋白酶(如HIV-1蛋白酶),阻断蛋白前体的水解(对感染性病毒颗粒的产生是必须的),从而阻止病毒复制[2][3]。
GW433908是一种有效的抗逆转录病毒蛋白酶抑制剂,比安普那韦(amprenavir)胶囊的溶解性更好。已有许多临床研究用于检测GE433908的安全性和药物动力学曲线。在健康男性志愿者中,GW433908以片剂或混悬液的方式给药,食物对其药代动力学有轻微影响,对片剂GW433908的影响更小。片剂GW433908耐受性良好,血浆中amprenavir的浓度与推荐的amprenavir治疗剂量相当,可用于治疗HIV感染[4]。作为抗逆转录病毒蛋白酶抑制剂amprenavir的磷酸酯前药,GW433908(1395 mg,QD)与efavirenz(200 mg,QD)联合给药可减少健康志愿者血浆中的APV暴露[1]。
